Organelle Isolation

Ultrasonic homogenization is a commonly used method for isolating subcellular fractions from various cell and tissue types. This technique works by exploiting the differences in size, shape, and density between organelles. By subjecting the sample to ultrasonic homogenization, it is possible to separate and purify specific organelle fractions. To be useful for further analysis, it is important for the isolated organelles to be pure, intact, and functional, which may require you to operate your ultrasonic homogenizer at lower amplitudes for longer periods of time. It is also important to confirm the integrity of the organelles using reliable assays.

At Qsonica, we offer a wide variety of Sonicators that are reliable, powerful, and proven. When you need a trustworthy instrument for tissue homogenization, do what tens of thousands of scientists have done for half a century: rely on a Sonicator.

Tips & Info for Organelle Isolation Application

  • Organelle isolation is a difficult procedure which requires careful protocol development. Be patient and give yourself plenty of test runs to optimize!
  • The goal is to do just enough to lyse a high percentage of the cells without damaging many of the desired organelles. Aim low when setting your amplitude and time.
  • A higher yield is possible when you want to extract smaller organelles. The greater the difference in size between your cells and the organelles you are looking to recover, the easier and more successful your organelle isolation process will be. On the other hand, isolating large organelles such as nuclei from small cells can be very difficult and result in relatively low yields.
  • Set realistic expectations. You are very unlikely to achieve anywhere near 100% yields when using any homogenization technology for organelle extraction.
  • For general best practices, see Best Practices for Ultrasonic Homogenization.
