Tissue Homogenization

Ultrasonic homogenizers make use of the ultrasonic waves generated by a piezoelectric crystal. When a certain voltage is applied to the crystal, the ultrasonic waves are transmitted through water or air, leading to cavitation which in turn leads to the generation of forces on nearby surfaces. These powerful forces homogenize tissues, breaking apart connective tissues , lysing cells, and releasing cellular contents which may then be purified and utilized in a wide range of downstream analyses.

Tips & Info for Tissue Homogenization

  • Sonicators are for research use only.
  • Sonicators are suitable for use with soft tissues. Hard tissues, such as bone or wood, are not suitable for use with ultrasonic homogenizers and may damage the horn.
  • Very tough, fibrous tissue such as ligament may not be homogenized efficiently using an ultrasonic homogenizer. Should you wish to process such tissue, you may wish to pre-treat with collagenase or a similar protease to soften the tissue.
  • All tissue being processed must be fully immersed in liquid, and the tip of the horn must remain below the liquid surface at all times.
  • Do not over-homogenize! Running for longer than necessary may degrade RNA, denature proteins, and shear DNA.
